Characterisation of trehalose-6-phosphate phosphatases from bacterial pathogens

چکیده
The trehalose biosynthesis pathway has recently received attention for therapeutic intervention combating infectious diseases caused by bacteria, helminths or fungi. Trehalose-6-phosphate phosphatase (TPP) is a key enzyme of the most common and particularly attractive target owing to toxicity accumulated trehalose-6-phosphate in pathogens.

A trehalose-6-phosphate synthase gene from Phellinus igniarius
Trehalose, an important disaccharide in fungal spores, plays important role in protects against many environmental stresses, such as heat and salt. In this study, a gene (PiTPS) encoded trehalose-6-phosphate synthase was isolated from Phellinus igniarius (L.: Fr.) Quél, a basidiomycete fungus belonging to the family Polyporaceae. It contains many bioactive compounds that have been reported to p...
متن کاملInteractions among sites responsible for trehalose 6-phosphate and trehalose-activated glucose 6-phosphate hydrolysis on trehalose phosphatase isolated from Phormia regina.
An enzyme capable of carrying out a trehalose-activated hydrolysis of glucose 6-phosphate has been purified 86-fold from whole body extracts of Phormia regina. By means of a number of criteria, it has been shown that the same enzyme is capable of activity against trehalose-6-P, although hydrolysis of the latter substrate is not activated by trehalose. It appears that the hydrolytic sites are ov...

Trehalose 6-phosphate (Tre6P), the intermediate of trehalose biosynthesis, is an essential signal metabolite in plants, linking growth and development to carbon status. The Suc-Tre6P nexus model postulates that Tre6P is both a signal and negative feedback regulator of Suc levels, forming part of a mechanism to maintain Suc levels within an optimal range and functionally comparable to the insuli...

T6P (trehalose 6-phosphate), the precursor of trehalose, has come out of obscurity over 10 years to be appreciated as an important regulator of plant metabolism and development, quite possibly linking the two. This information has been gained from analysis of mutant and transgenic plants, which show strong, diverse and strategically important phenotypes. Plant genes that encode the trehalose pa...
متن کاملDisruption of TPS2, the gene encoding the 100-kDa subunit of the trehalose-6-phosphate synthase/phosphatase complex in Saccharomyces cerevisiae, causes accumulation of trehalose-6-phosphate and loss of trehalose-6-phosphate phosphatase activity.
Preparations of the trehalose-6-phosphate synthase/phosphatase complex from Saccharomyces cerevisiae contain three polypeptides with molecular masses 56, 100 and 130 kDa, respectively. Recently, we have cloned the gene for the 56-kDa subunit of this complex (TPS1) and found it to be identical with CIF1, a gene essential for growth on glucose and for the activity of trehalose-6-phosphate synthas...
